I feel your pain. I have been down this road too, all I can say is don't give up and realize even a failure can be very valuable information for the future. You won't know if something works until you try it.

I think your next step with the head upgrade will solve the detonation problem.
Also are you measuring the E85 alcohol strength. I have been burned a few times with pump E85 being as low as E65. It cost me a engine.

Anyway keep us informed on how it goes.

I have a '95 F-150 with a 4.9 (300) 6 cylinder, last year of the OBD-1. My ultimate goal is to build a gasifier and to run it on woodgas. But, I also want to run it on E85 or E100. Both ethanol and woodgas call for higher compression and advanced timing. But, I don't know beyond that.

I'm thinking the 4.9 is multi-port fuel injected rather than a single throttle body. Just haven't spent much time under the hood. A local machine shop is interested in this build. They do mostly race engines but like the idea of this research project.

Suggestions? I'm thinking larger injectors. What about cam? Computer? I've even considered going to a MegaSquirt. But, I want it to be a daily driver using pump E85.

You can start doing that right now by simply filling with E85.

Before you try E85 in that F-150, you'd be better off replacing all the fuel lines to prevent them from get damaged by the ethanol.
